Here's my idea for a Marvel Cinematic Universe take on the Sinister Six. Basically MCU's equivalent of the DCEU's Suicide Squad.
    J. Jonah Jameson: Well known Anti-Spider-Man Advocate, The main protagonist of the film, portrayed by J.K. Simmons. The main plot sees Jameson's stubborn arrogance in proving that Spider-Man is NOT a real hero get him in big trouble as he finds himself the leader of this film's incarnation of the Sinister Six.
Otto Octavius / Doctor Octopus: A misguided scientist who uses his arm harness to conduct an Oscorp-funded fusion experiment using tritium that becomes impossible to control, resulting in the fusion of the harness to Octavius's body and the destruction of the inhibitor chip that keeps the arms' artificial intelligence in check. Similarly to Triple J, Doc Ock is once again portrayed by Alfred Molina (Spider-Man 2).
Flint Marko / The Sandman: Once a small time crook who stole to pay for medical treatment for his critically ill daughter, Penny. While on the run from the police after escaping from prison, he accidentally falls into an experimental particle accelerator that binds him with sand on a molecular level, giving him shapeshifting sand abilities & transforming him into the Sandman. Similarly to Triple J & Doc Ock, Sandman is once again portrayed by Thomas Haden Church (Spider-Man 3), & his part in the story revolves around his dynamic with Spider-Man, wanting Peter to understand that his love for his daughter Penny is the only thing that drives him to commit crime.
Felicia Hardy / Black Cat: The daughter of Walter Hardy, a world-renowned cat burglar, finally making her live-action debut with either Anne Hathaway or Felicity Jones portraying her. After suffering a traumatic experience as a college freshman, Felicia, who hates the idea of being a victim, trains herself in various fighting styles and acrobatics, deciding to follow in her father's footsteps with intent on getting revenge against those who wronged her. As Black Cat, she has the subconscious ability to affect probability fields, producing "bad luck" for her enemies.
Adrian Toomes / The Vulture: Having already debuted in the MCU as the main antagonist of Spider-Man: Homecoming, The Vulture gains a new suit.
Once again portrayed by Michael Keaton.
Herman Schultz / Shocker: Having already debuted in Spider-Man: Homecoming as a henchman of The Vulture, Schultz gains a new set of gauntlets... and a familiar looking suit to match.
Once again portrayed by Bokeem Woodbine.
And for the film's main villain... Norman Osborn / Green Goblin: With Willem Dafoe reprising his role from Sam Raimi's original Spider-Man film, Norman Osborn is the strong-minded yet troubled head of Oscorp who has a distant relationship with his son while possessing a deep respect & admiration for the Avengers. The Green Goblin is depicted as an alternate personality, separate from Osborn who acts as the unhinged enforcer of Norman's desires, claiming that Norman is too weak to achieve them himself. Problems arise for Osborn when he faces the threat of losing a military contract should an attempt at replicating the super soldier formula that created Captain America not receive proper human testing. As the loss of military funding would likely mean the end of his company, the desperate Osborn tests the formula on himself which simultaneously grants him super strength & drives him insane. Upon transforming into The Goblin, the delirious Norman steals Oscorp's experimental one-man flight vehicle known as "The Glider" along with the cybernetic flight suit that comes with it. Forcing J. Jonah Jameson to take up the role of Sinister Six leader as part of a Xanatos Gambit that would result in either Spider-Man abandoning the path of the hero & joining HIS side or Spider-Man being destroyed, The Green Goblin attacks with his glider & pumpkin bombs that upon detonation, can vaporize anything caught within blast radius.
And his accomplice... returning from Spider-Man: Homecoming... Mac Gargan / Scorpion: Initially a normal mobster, Mac Gargan makes his debut as the super-powered Scorpion we're all familiar with. Disgraced by Spider-Man, Gargan was hired by Jameson as the subject of a barely-tested process that would endow him with the useful characteristics of a particular animal with the help of Dr. Farley Stillwell. Dr. Stillwell also outfits him with a club-like mechanical tail. In this case, a scorpion is the base for the purposes of creating a powerful agent capable of defeating Spider-Man as the scorpion is a natural predator of the spider. Unfortunately for Triple J, the mutagenic treatment seriously affects Gargan's mind, driving him insane as the predatory instincts of the scorpion take over. Promptly turning on Jameson, The Scorpion becomes a professional criminal to indulge in a vendetta both against Jameson, & against Spider-Man for foiling his attempt to kill him. He also turns on his former associate, The Vulture, for refusing to give him information on Spider-Man's secret identity, which he had already learned by then thanks to Mysterio exposing the web-head in the first post-credits scene of Spider-Man: Far From Home
Aside from The Friendly Neighborhood Spider-Man himself, I'm also thinking of the following Spider-Man alumni making appearances...
Vincent D'Onofrio as Wilson Fisk / The Kingpin
Paul Giamatti as Aleksei Sytsevich / The Rhino
Jamie Foxx as Maxwell "Max" Dillon / Electro
Stephen Oyoung as Martin Li / Mister Negative
Jake Gyllenhaal as Quentin Beck / Mysterio

This REALLY makes me hope for the sequel to Spider-Man on PS4, they'll add the underarm webs as an ability to glide through the city, that would be amazing!

This is what we all wanted to see. A brand new Spiderman taken out from the comics, swinging around New York, in the same universe with the Avengers. In my opinion, this is probably my favourite scene of Spiderman as it represents the character very well.
